The show was originally developed as Looney Tunes Laff Riot, a 'true-to-the-classics' project announced in July 2009 that aimed to emulate the original theatrical shorts, but it was scrapped after executives were unimpressed and later retooled into the sitcom-style The Looney Tunes Show.

The Laff Riot pilot, which featured a duck character named Marisol Mallard (later renamed Tina Russo), remained unreleased for over a decade until it surfaced online on September 4, 2020.

Character designer Jessica Borutski, based in Ottawa, created new designs for the Laff Riot pilot which were later retooled and reused for the final series.

Actress Kristen Wiig voiced Lola Bunny for the series, but she was replaced by series writer Rachel Ramras for the direct-to-video film Looney Tunes: Rabbits Run.

The character Witch Lezah, Gossamer's mother, is based on Witch Hazel, with her name simply being 'Hazel' spelled backwards; promotional artwork by Jamie Nicholes even depicted Lezah alongside Hazel, who was originally planned to appear in the series.

Yosemite Sam is given the full name Samuel 'Yosemite Sam' Rosenbaum in the show, portrayed as far less ruthless and more mild-mannered than in the classic shorts.

When the redesigned Looney Tunes characters were first revealed in 2010, some fans reacted with hostile online comments directed at designer Jessica Borutski, who said it was hard to see such hatred but defended the redesigns as a way for a new generation to meet the characters.

This series marked the final time June Foray voiced Granny before her death in 2017.
